Description:
MAP3K7 is a serine/threonine kinase member of the mitogen-activated kinase kinase kinase (MAP3K) family, which forms part of a three-tiered core-signalling module (MAP3K→MAP2K→MAPK), a defining feature of MAPK pathways. MAP3K7 is a key signalling component of extracellular signal-regulated kinase (Erk) 1/2, nuclear factor-kappa B (NF-kB), p38 MAPK and c-Jun N-terminal kinase (JNK) pathways. MAP3K7 responds to diverse stimuli, including transforming growth factor-beta (TGFβ)-related agents, proinflammatory cytokines such as TNF-α and IL-1β, TLR ligands such as lipopolysaccharides, and various types of stress signals. The transmission of signal is mediated by sequential phosphorylation and activation of kinases in the MAPK modules, which work together with scaffold proteins and other regulators to impart specific and unique kinetic properties in a context-dependent manner. MAP3K7 functions in the regulation of immune response, survival, apoptosis, development, differentiation and carcinogenesis. Polymorphisms in this gene associated with HbF levels in sickle cell patients. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.
