Description:
The encoded protein is a potent pro-inflammatory cytokine, part of the tumor necrosis factor (TNF) superfamily, derived predominantly from activated macrophages. It acts on various signalling pathways through specific cell surface receptors, TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR to regulate NF-kB activation of inflammation, stress-activated protein kinases, and apoptotic pathways. It functions mainly in the containment of local infection and is involved in the regulation of several biological processes, including cell proliferation, differentiation, apoptosis, lipid metabolism and coagulation. It has also been implicated in noninfectious inflammatory diseases, including rheumatoid arthritis, osteoarthritis, atheroschlerosis, insulin resistance, Alzheimer's disease and cancer. Polymorphisms in this gene associated with stroke in sickle cell patients.
