Description:
The protein encoded by this gene belongs to the lin-28 family, which is characterized by the presence of a cold-shock domain and a pair of retroviral-type CCHC zinc finger domains. It is highly expressed in the testis, fetal liver, and placenta. LIN28B binds and inhibits let-7 miRNAs, which function as important regulators of fetal-to-adult developmental transition in erythroid tissues. Overexpression of LIN28B proteins or decreased expression of its target let-7 miRNAs enhanced fetal haemoglobin (HbF) levels in adult erythroid cells and ameliorated the hypoxia-related sickling of cultured erythroblasts. Independently of its role in regulating let-7 microRNAs, LIN28B directly interacts with and suppresses BCL11A, a key regulator of developmental haemoglobin switching and silencing of HbF in adults. LIN28B is a candidate regulator of HbF expression.
