Description:
The Duffy antigen receptor for chemokines (DARC/ACKR1) belongs to the family of atypical chemokine receptors (ACKRs) that bind to a variety of proinflammatory chemokines without eliciting a biological signal. Members of this family share the seven-transmembrane α-helix structure of G protein-coupled receptors, but lack the entire DRYLAIV sequence at the end of the third transmembrane domain, which is required for G-protein coupling and subsequent signalling. DARC is present on erythrocytes and post-capillary endothelial cells, as well as on a subset of neurons such as cerebellar Purkinje cells. DARC appears to regulate chemokine bioavailability and, consequently, leukocyte recruitment that helps reduce inflammation by scavenging chemokines in the circulation. DARC also serves as the erythroid receptor for the malarial parasites Plasmodium vivax and Plasmodium knowlesi. Two transcript variants encoding different isoforms have been found for this gene. Polymorphisms in this gene associated with pulmonary hypertension.
