Description:
This gene encodes E-selectin, an adhesion molecule expressed on endothelium in response to inflammatory stimulation. It is a member of the family of C-type lectins, which are involved in leukocyte adhesion and rolling on the vascular endothelium. E-selectin binds to glycoproteins containing the tetrassacharide sialyl Lewis x (sLE x) in a calcium-dependent manner. Compared to other selectins, E-selectin has a diverse set of ligands and enables slow rolling on the order of a micron per second. Rolling transduces signals that trigger β2 integrin activation, which leads to firm leukocyte arrest and transmigration to sites of injury. E-selectin co-operates with the chemokine receptor CXCR-2 to facilitate this process. It plays a role in inflammation. It has an important role during haematopoietic stem/progenitor cell trafficking to the bone marrow niche. A polymoprhism in this gene presents a risk factor of venous thrombosis in patients with HbH disease.
