Description:
NFIX belongs to the nuclear factor 1 (NF1) family of transcription factors that also includes NFIA, NFIB and NFIC. NFI proteins consist of a conserved N-terminal DNA-binding and dimerization domain and a C-terminal transactivation/repression domain. They exhibit a high variability due to extensive alternative splicing. NFIX is highly expressed by haematopoietic stem and progenitor cells (HSPCs) and plays an important role in regulating their repopulating potential and survival. It has been shown to regulate myeloid-vs-lymphoid cell fate at early stages of HSPC development, potentially via the direct modulation of lineage-specific transcription factors. NFIX and NFIA are the predominantly expressed NIF forms in human erythroid cells, expressed at higher levels in adult compared to fetal erythroid cells. Both NFIX and NFIA are involved in the silencing of the gamma-globin genes. The disruption of NFIX has little effect on HBG1/2 expression, whereas the combined disruption of NFIA and NFIX, as well as the disruption of NFI binding sites near the HBG1/2 genes produced a higher, albeit comparable, increase in HBG1/2 expression. The NFIX gene is a candiadte regulator of HbF expression.
