Description:
MicroRNAs (miRNAs), such as MIR96, are small, non-coding RNAs that play key roles in the post-transcriptional regulation of gene expression by targeting specific mRNAs. Reporter gene assays demonstrated that MIR96 directly and sequence specifically targets the open reading frame (ORF) of the γ-globin mRNA, thereby inhibiting γ-globin expression. Additional evidence showed that MIR96 expression levels inversely correlated with increased HbF synthesis in erythroid cells of beta-thalassaemia patients and after hydroxyurea treatment in sickle cell anaemia patients. MIR96 acts as a negative regulator of γ-globin gene. Therapeutic approaches that increase the HbF concentration have great potential for ameliorating the clinical and haematologic severity of beta-thalassaemia and other beta-haemoglobinopathies.
